Your question is very broad and it's a minimum of 6 months courses to study not an answer to receive with a single question as you thought!

In my opinion, it's better to use frameworks like flask or django for python. So it's easier to make projects that you want. Also I think that JS and Python should be both considered as back - end. And JS is more like full stack, i.e. you can use it on both ends. But basicaly, you use "front-end" lang to communicate to your "back-end" some instructions. And then back-end does what it does and gives feedback. Etc.
