There are many ways, but I find Tkinter the best. Firstly, run Python 3.5 or whichever you have. Type into the command line "import tkinter". After that, type " tkinter._test()". It should open a small window, where you just press the "Quit" button. Now you can programme the GUI. You can do it in IDLE as well, since IDLE itself was written in Tkinter.
