What are the main initial topics a Python beginner should perfectly know?

The Introduction to Python course should give you a good idea about the very basics, and you have already completed that one.

I don't know about "perfectly". I would go for "sufficiently enough to be usable".
Perfect knowledge will have some nasty surprises for you, your world will crumble a few times when you get more advanced with Python. :)
